摘  要:目的建立测定全营养混合液(TNA)中黄芪甲苷含量的方法。方法采用高效液相色谱-蒸发光散射检测(HPLC-ELSD)法对TNA中黄芪甲苷的含量进行测定,色谱柱为Kromasil C18柱(250mm×4.6mm,5μm),流动相为乙腈-水(35∶65),流速为1.0mL/min,ELSD参数为漂移管温度105℃,气体流速2.8L/min。结果黄芪甲苷对照品进样量在2.10~10.50μg范围内与峰面积线性关系良好(r=0.9952),平均回收率为96.96%,RSD=1.69%(n=9)。结论HPLC-ELSD法操作简便、结果准确、重现性好,可作为该制剂中黄芪甲苷含量的检测方法。Objective To establish the assaying method of astragaloside Ⅳ in total nutrient admixture (TNA). Methods To determine astragaloside IV in TNA by HPLC- ELSD. Kromasil C18 column (250 mm ×4. 6 mm, 5 μm) was used. The mobile phase was acetonitrile- water (35:65).The flow rate was at 1.0 mL/min. The drift tube temperature was at 105 ℃ and the gas flow rate was 2.8 L/min. Results A good linear relationship between the content and peak area of astragaloside IV was found within the range of 2. 10- 10. 50 μg. The regression coefficient was 0. 995 2 and the average recovery rate was 96.96%, RSD was 1.69% (n =9). Conclusion The method is convenient, accurate with fine reproducibility. It can be used for the quality control of astragaloside Ⅳ.
