槐耳颗粒对小鼠心脏移植物生存期的影响

摘　　要：目的探讨槐耳颗粒对小鼠心脏移植后移植物生存期的影响及其可能机制。方法槐耳颗粒在小鼠心脏移植术后每天予以6mg/g体重槐耳清膏灌胃处理;移植排斥对照组和同系移植对照组灌清水;术后5d或移植排斥终点收集供心做石蜡病理切片行HE染色及免疫荧光检查。结果移植排斥对照组供心平均存活时间为(8.0±0.53)d,槐耳颗粒组供心平均存活时间为(6.0±0.26)d,两组差异有统计学意义(P<0.001)。槐耳颗粒组和移植排斥对照组供心心肌变性坏死、大量CD8+T细胞浸润及FasL表达。而同系移植对照组阴性。结论小鼠心脏移植术后槐耳清膏灌胃移植物生存期缩短,其可能与其增强CD8+T细胞浸润及FasL表达有关。Objective To investigate the effects and its potential mechanisms of trametesrobiniophila murr granules on survival time of mouse heart grafts. Methods After the mouse heart transplantation, intragastric administration was done on the recipients with Chinese medicine extractum of trametes robiniophila murr （6 mg/g）. The control groups of allotransplantation and syngenic transplantation were treated by intragastric administration of purified water. The donor＇s heart was harvested at 5th day after operation or at the end of the graft rejection and embedded by paraffin, and cut into sections stained with hematoxylin and eosin and immunofluorescence. Results The average survival time of the grafts was 8.0 ±0.53 days in the allotransplantation group and 6.0 ±0.26 days in the group of rametes robiniophila murr granules, respectively, with the difference being statistically significant （ P 〈 0. 001 ）. There were degenerations and necrosis, infiltration of CD8^＋ T cells and expression of Fas ligand in cardiac muscle tissues in these two groups. But the syngenic transplantation group differed in all above aspects contrast to the two groups. Conclusion During mouse heart transplantation, the survival time of grafts is shortened by intragastric administration of Chinese medicine extractum of trametes robiniophila mutt, which was probably related to the enhanced infiltration of CD8^＋ T cells and expression of Fas ligand.
