摘 要:用反相制备液相色谱法从辣椒碱类化合物中分离得到了高纯度辣椒碱单体。采用碳十八硅胶(ODS)柱(200mm×10.0mm),考察了流动相组成、进样浓度、进样体积和流动相流速等对分离的影响,确定了适宜的工艺操作条件:流动相为甲醇/水(75/25,v/v),进样浓度为180mg·mL-1,进样体积为2.08mL,流速为4.0mL·min-1。在该工艺条件下,得到的辣椒碱单体纯度为95.23%,回收率为93.27%,产率为4.38g·L-1·h-1,流动相消耗为0.087L·g-1。本工艺操作简便,分离效率高,产品质量好,为辣椒碱单体的制备研究提供了一种新的方法。Capscaicin was separated from capsaicinoide by reversed-phase preparative liquid chromatography. Using a ODS column (200 mm× 10.0 mm), the effects of mobile phase and flow rate, feed concentration and volume on the purity, productivity, recovery and mobile phase consumption were studied respectively, and the suitable operation conditions were obtained as follows: the mobile phase is methanol/water (75/25 v/v), and the feed concentration and volume are 180 mg.mL^-1 and 2.08 mL, respectively; while the flow rate is 4 mL.min^-1. Under above conditions, the capscaicin with a purity of 95.23% could be achieved, and the productivity, recovery and mobile phase consumption are 4.38 g.L^-1.h^-1, 93.27% and 0.087 L.g^-1, respectively. This technology proposed provides a simple and effective method for the preparation of capscaicin.
