摘  要:观察成年期甲状腺功能减退症(简称甲减)大鼠海马突触蛋白synaptotagmin 1(syt 1)改变及替代治疗和冲击治疗的影响。结果显示,甲减大鼠血清T3及海马CA1、CA3区和齿状回(dentate gyrus,DG)syt 1水平显著性降低,替代治疗后血清T3恢复正常,但syt 1水平仅DG分子层显著性增加,而经冲击治疗后血清T3、T4高于正常,海马CA1区和DG区各层及CA3区锥体细胞层syt 1的水平几乎均恢复正常。这些结果提示,成年期甲减可致海马内syt 1蛋白表达减少,甲状腺素治疗能使其恢复,短期冲击治疗较替代治疗效果好。We observed the expression of synaptotagmin-1(syt 1) in hippocampus of adult hypothyroid rats and the effects of replacement or shock therapy by thyroid hormone on syt 1.Our results showed that the serum T3 level and syt 1 immunoreactivity in all layers of CA1,CA3 and DG regions of hippocampus in the hypothyroid rats decreased significantly.After replacement therapy of thyroid hormone,the serum T3 level returned to normality,but the significant recovery of syt 1 immunoreactivity occurred only in molecular layer of DG.After shock treatment of thyroid hormone,serum T3 and T4 levels were higher than the control group,all layers of each subfield had significant recovery of syt 1 immunoreactivity,with similar level to the control group.These results suggested that the expression of syt 1 decreased in CA1,CA3 and DG regions of hippocampus in adult-onset hypothyroid rats,but this decrement could be reversed by supplement of thyroid hormone,and the effect of L-thyroxine shock treatment is superior to that of replacement therapy.
