冬夏两季畜禽废水的活性污泥中细菌遗传多态性研究

摘  要:对冬夏两季膜生物器处理畜禽废水的效果进行研究,分析两季废水中细菌遗传多态性与废水处理效果的关系。直接提取废水中活性污泥总DNA,构建16S rDNA基因文库。选取两季各8个成功的阳性克隆子,分别采用HinfⅠ、HaeⅢ、AluⅠ、M spⅠ4种限制性内切酶消化重组质粒,电泳分别表现出不同谱型。结果表明,4种限制性酶中,HinfⅠ的酶切图谱反应的遗传信息量最丰富。细菌遗传多态性Shannon-Wiener指数冬季为0.953,夏季为2.079,即夏季多态性高于冬季;根据出水水质监测,夏季处理效果优于冬季,与多态性指数呈正相关。By using MBR reactor to probe the different effects of livestock wastewater's treatment in winter and summer,the relationship between the microbe genetic polymorphism and treatment efficiency was analyzed. We extracted the total DNA from activated sludge to construct the 16S rDNA gene-library by cloning, then picked out 8 white spotting respectively and extracted plasmids of them. The plasmids of recons were digested at 37℃ by HinfI , Hae Ⅲ, Alu Ⅰ and Msp Ⅰ ,respectively. The result showed that HinfⅠ produced the richest gene diversity index. The Shannon-Wiener information index was O. 953 in winter and 2. 079 in summer. Thus,the summer's outlet was much better than the winter's, that means the genetic diversities in different seasons were related to their water qualities of outlet.
