银强化滴金免疫法检测丙肝病毒IgM的研究

摘　　要：目的建立一种检测血清丙肝病毒抗体IgM的方法。方法将丙肝病毒抗原点于硝基纤维膜上,应用Φ15nm的羊抗人IgM金标抗体与血清标本中的丙肝病毒IgM结合,再加银加强试剂,阳性者为棕黑色。结果整个试验过程需要10m in,检测HCVAb/HCVRNA阳性临床标本,结果有良好的一致性,总符合率为96%。结论银强化滴金免疫法检测血清丙肝病毒抗体IgM,操作简单,试剂稳定、敏感、快速,适于基层。Objective To establish a more rapid and simple method for detecting anti - HCV IgM antibey tin patient＇ s serum. Methods A silver enhanced clot immunogold filtration assay（SEDIGFA） was developed by coating the antigen protein of HCV on nitrocellulose （NC） membranee, adding colloidal gold labeled sheep anti - human IgM antibody and the silver enhancer. The whole procedure can be finished within 10 minutes. Results The coincident rate of serum samples positive for HCVAb/HCVRNA detected with this method was 96% . Conclusion SEDIGFA is more simple and it can be used for rapid detecttlon of anti - HCV IgM in small medical laboratories.
