摘  要:目的探讨不同发育阶段大鼠睾丸内c-kit的表达特点。方法采用RT-PCR、免疫组化、Western blot方法分别检测1日龄、9日龄、1月龄大鼠睾丸内c-kit mRNA和蛋白的表达,并进行半定量分析。结果1日龄、9日龄和1月龄大鼠睾丸内均有c-kit mRNA和蛋白的表达,9日龄组大鼠睾丸内c-kit mRNA表达水平(0.2040±0.0048)和蛋白表达水平(3.7263±0.0087)显著高于1日龄组(0.0825±0.0071,2.4270±0.0982)和1月龄组(0.1034±0.0064,2.6532±0.1150)大鼠(均P<0.01),1日龄组与1月龄组c-kitmRNA和蛋白表达水平差异均无统计学意义(均P>0.05)。结论c-kit的表达与精原细胞分化有直接关系,c-kit是精原细胞走向分化的标志。Objective To investigate the c-kit expression pattern in rat testes of different developmental stages. Methods RT-PCR, immunohistochemistry and Western blot were performed to detect c-kit mRNA and protein expression in 1-day-old, 9-day-old and 1-month-old rat testes. All results were subjected to semiquantitative analysis. Results C-kit mRNA and protein expression was detected in 1-day-old, 9-day-old and 1-month-old rat testes. In 9-day-old rat testes, the expression of c-kit mR- NA and protein was notably higher than in 1-day-old and 1-month-old rat testes. There was no significant difference between 1- day-old and 1-month-old rat testes in the c-kit expression. Conclusion Expression of c-kit has a direct relationship with differentiation of spermatogonia. C-kit is a signal of spermatogonia which undertake differentiation.
