摘  要:目的考察用合煎(传统汤剂)、分煎(配方颗粒汤剂)方法制备的鸭掌散汤剂中麻黄碱含量的变化,并比较其体外抗菌作用。方法①建立高效液相色谱方法,采用Diamonisl C18柱(250 mm×4.6mm,5μm),乙腈-0.1%磷酸(5∶95)为流动相,流速1ml/min,检测波长207 nm。分析鸭掌散合煎液和分煎液样品中麻黄碱的含量变化。②采用稀释法体外测试鸭掌散不同煎液对常见菌种的最低抑菌浓度(MIC)和最低杀菌浓度(MBC)。结果①鸭掌散分煎液中麻黄碱的平均含量大于合煎液中麻黄碱的平均含量。②鸭掌散分煎液对福氏痢疾杆菌有抑菌作用,而合煎液没有;合煎液对白色葡萄球菌、黄曲霉菌的抑菌作用大于分煎液。结论与传统汤剂比较,鸭掌散配方颗粒汤剂中麻黄碱的含量较高,抗菌谱较广。Objective To investigate the contents of ephedrine hydrochloride in combined decoction ( mix the traditional Chinese herbs together, then boil them with water) and the separated decoction (boil the single traditional Chinese herb with water separately, then mix the abstracts) of Ya Zhang powder ( composed of Ephedra sinica, Prunus armeniaca and Glycyrrhiza uralensis ) and to compare their anti - bacterial effects in vitro. Methods ①An HPLC method was established with a Diamonisl C18 column (250 mm ×4.6 mm,5μm) and mobile phase of methanol-0.1% phosphoric acid(5: 95). The flow rate was 1ml · min^-1 and the detection wavelength was 207nm. ②Minimal bacterial inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) against the common bacteria in different decoctions were measured in vitro by employing dilution method. Results ①The average content of ephedrine hydrochloride in the separated decoction was higher than that of the combined decoction. ②The separated decoction had the antibacterial effects to virulence of shigella flexneri, but the combined decoction did not ; And to Staphylococcus albus and aspergillus flavus, the antibacterial effect of combined decoction was slightly stronger than that of the separated decoction. Conclusion Compared with that of the separated decoction, the content of ephedrine hydrochloride in the combined decoction was higher and the anti -bacterial effect was stronger.
