摘  要:目的研究短尾蝮蛇毒磷脂结合抗凝蛋白体内外的抗凝作用及机制。方法按试剂盒说明进行磷脂结合抗凝蛋白对活化部分凝血活酶时间(APTT)、凝血酶原时间(PT)、凝血酶时间(TT)、纤维蛋白原(FIB)含量、凝血因子活性、组织凝血活酶抑制实验(TTI)和稀释的罗氏蝰蛇毒时间(DRVVT)、血小板聚集功能以及溶血、出血实验。结果磷脂结合抗凝蛋白能显著地延长活化部分凝血活酶时间和稀释的罗氏蝰蛇毒时间,减少组织凝血活酶抑制实验(TTI)试剂的磷脂浓度,能加速其抗凝作用,干扰Ⅷ,Ⅸ,Ⅺ,Ⅻ因子活性,未见溶血和出血。其抗凝活性与磷脂结合有关,可能通过内源性途径抑制凝血过程。结论短尾蝮蛇毒磷脂结合抗凝蛋白能够抗凝、抑制Ⅷ,Ⅸ,Ⅺ,Ⅻ因子活性并与磷脂结合,可通过内源性途径抑制凝血过程。Objective To study anticoagulation effects of Phospholipid -binding anticoagulation protein(PBAP) from Agkistrodon halys Brevicaudus Venom. Methods The activated partial thromboplastin time (APTF), Prothrombin time (PT), Fibrinogen (FIB), Thrombin time(TT) and Factor activity were determined by kit illustration. The TTI test was peformed according to the method of Schleider . The dRVVT test was done in quadruplicate using commercial kits acoording to the manufacturer's iustrution. Results Comored with conred group,the Phospholipid -binding anticoagulation protein(PBAP) from Agkistrodon halys Brevicaudus Venom could markedly prolong APTT and DRVVT( P 〈 0.01 ) concentration -dependent but time -independent manner, accentuate the inhibitor effect of TTI by decreasing phospholipid concentration, and inhibit the activities of FⅦ : C, FⅨ : C, FⅪ : C, FⅫ :C. Conclusion The Phospholipid - binding anticoagulation protein from the Agkistrodon halys Brevicaudus Venom can anticoagulate and inhibit FⅧ : C, FⅨ : C, FⅪ : C, FⅫ : C activity and bind Phospholipid.
