摘  要:目的观察中药脑宁康(NNK)颗粒对经脑缺血预适应(IPC)处理后大鼠HSP70表达的影响。方法根据Pulsinelli四管法及Liu法分别制作大鼠IPC及脑缺血再灌注(IR)模型,应用免疫组化SP法作HSP70染色,比较NNK颗粒对IPC和IR两种模型大鼠HSP70表达的影响,并以阿斯匹林作对照。结果NNK在两模型中均可使HSP70表达明显增强(P〈0.05~0.01),且效应显著高于阿斯匹林(P〈0.05),但两模型之间比较无显著性差异。且NNK能够延缓HSP70表达强度的减弱。结论NNK颗粒通过增强IPC和缺血状态中HSP70表达,扮演了强化IPC及药物预适应角色,旨在增加脑神经细胞对缺血、缺氧的耐受力,并通过对HSP70的干扰,获得稳定神经细胞结构,维护其正常功能的药理效应。Objective To investigate the effect of Naoningkang particle on the expression of the 70KDa heat shock protein (HSP70) following the ratslschemic precondition . Methods The rats'IPC and ischemic reperfusion ( IR ) models were made by methods of Pulsinelli four tubes and Liu respectively. The expression of HSP70in two rat models including IPC and IR were compared by NNK particle applying SP immunohistochemistry method. Results NNK particle could increase the HSP70 expression of IPC and in the state of ischemia and play the role on stronging ischemic precondition of IPC and medicine. The effect of NNK particle suggests increasing the tolerance of central nerve cells facing to ischemia and hypoxia. Conclusion NNK particle also can stabilize the strueture of neuron through interfere with HSP70 expression , and then it demonstrate the pharmacologic effect of keeping the neuron normal functions.
