摘  要:目的探讨G-CSF在应用于干细胞动员治疗心血管疾病时对血小板功能的影响。方法16只新西兰大白兔随机分为普通和高脂喂养2组,予G-CSF皮下注射5日,用药前后分别测定PLT、MPV、PDW、P-LCR、TXB_2、P选择素等血小板相关指标,并采用全血CD41-FITC/CD62P、CD63-PE荧光抗体双标流式分析法进行验证。结果用药后普通喂养组除PLT计数外上述各项指标与基础状态相比均增高。但高脂喂养组除P-LCR外,其他指标无显著变化。而用药前后者的P-LCR、TXB_2高于前者。结论G-CSF在作用高峰能引发血小板功能的活化,其机制可能与应激反应有关。高血脂本身就有激活血小板的作用。全血细胞流式分析是检测血小板活化的较灵敏方法。Objective To study the effect on platelet function by G-CSF used to be stem cell mobilization for treating cardiovascular disease. Methods 16 New Zeeland healthy male rabbits were divided into 2 groups consisted of high fat feed and ordinary feed randomly,all the animals were given hypodermic injection of G-CSF for 5 days, and the indexes related to platelet function such as PLT, MPV, PDW, P-LCR, TXB2 ,P-selectin were tested before and after the drug injection. Finally those results were confirmed by the whole blood FCM symbolized by the CD62P and CD63 expressing rate, CD41 was used to set the blood platelet gate. Results After the injections all the indexes increased except the PLT counts in ordinary fed group, and the difference has statistics significance ( P 〈 0.05 ). The whole blood FCM also supported it. However all the indexes in high fat fed group hadnt remarkable changes except P-LCR,and the value of this index and TXB2 were higher than ordinary fed groupg before the drug injection. Conclusions G-CSF can induce the activation of platelet function when it get the mobilization peak, so the mechanism maybe relate to stress reaction. High blood fat level itself can activate the platelet function. The whole blood FCM is a good way to confirm it added to the results of ELISA and so on, but the essay technique need improvement to test bulk simultaneously.
