摘 要:目的建立引起该细胞增殖的方法,为以后进一步研究染毒后细胞内结构与功能的变化提供方法基础。方法用苯醌对人早幼细胞系HL-60进行染毒,取对数生长期的HL-60细胞,用不同浓度的苯醌进行染毒,行台盼蓝拒染法测细胞生存率,Alamar blue法测细胞增殖率,流式细胞技术测细胞周期分布。结果苯醌浓度为3μmol/L的剂量组HL-60细胞存活率无明显改变,且增殖率最高,细胞进入S期的比例[(48.23±1.37)%]高于对照组[(42.47±0.45)%]。结论用3μmol/L的苯醌染毒HL-60细胞,可引起该细胞明显增殖。Objective To establish a method for proliferation of HL-60 cells for further research on the structural and func- tional changes of these cells. Methods HL-60 was treated with different concentrations of benzoquinone (BQ). For testing cell viability, BQ-treated ceils were stained with trypan blue dye and counted. For proliferation evaluation, cells were stained with alamar blue dye and counted. HL-60 cells were also analyzed for their cell cycle distribution with a FACS sort flow eytometer. Results 3 μmol/L BQ did not significantly reduce cell viability as compared with the control cells, and the 3 μmol/L BQ-treated cell population showed the highest growth rate. The proportion of cells in S-phase at 3 μmol/L BQ- treated culture was higher than that of the control. Conclusion 3μmol/L BQ-treated HL-60 cells showed increased growth over the untreated control.
