四种鬼臼毒衍生物的体外抗肿瘤作用

摘　　要：目的：研究四种鬼臼毒衍生物在体外对Ｌ＿（１２１０）细胞生长的抑制作用。方法：采用活细胞计数法和软琼脂克隆形成法及有丝分裂指数计数法。结果：ＶＰ＿（１６），ＧＰ＿１，ＧＰ＿１Ｈ和ＧＰ＿１ＯＨ体外剂量，时间依赖性的抑制Ｌ＿（１２１０）细胞生长，２４ｈＩＧ＿（５０）依次为０．０４９，１．０６，３．８９，５．３３μｇ／ｍｌ，不同时间ＶＰ１６均降低细胞有丝分裂指数而ＧＰ１等反之。结论：ＶＰ＿（１６），ＧＰ＿１，ＧＰ＿１Ｈ和ＧＰ＿１ＯＨ的抗肿瘤机理有所不同。Objective: To study antitumor action of four derivatives of podophyllotoxin on L_(1210) cells.Methods: Inhibition ofproliferation were measured by trypan blue esclusion, colony-forming effeciticy and nutotic index. Results: The proliferationand colony formation of L_(1210) cells were sigrnificantly inhibited with dose and time-depedently by four derivatives ofpodophyllotoxin.In vitro. 24h IC_(50) of VP_(16), GP_1, GP_1H, GP_1OH were 0.049,1. 06, 3. 89, 5.33μg/ml, VP_(16) decieased themitotic index of L_(1210) cells but anther three drugs inereased it.Conclusion:four derivatives of podophyllotoxin all had antitu-mor effect on L_(1210) cell.
