摘 要:目的:建立大黄黄连泻心汤中君药大黄的主要蒽醌类成分(芦荟大黄素、大黄酸、大黄素、大黄酚和大黄素甲醚)的HPLC含量测定方法,并探索在不同配伍情况下浸渍剂中5个成分的含量变化。方法:采用HPLC-UV法,色谱柱为苯基柱(250mm×4.6mm,5μm),柱温为室温,流动相为甲醇- 0.1%磷酸水,梯度洗脱,流速为为1mL/min;紫外检测波长为254nm,进样量为20μL。结果:大黄的5个主要蒽醌类成分中的芦荟大黄素、大黄酸、大黄素、大黄酚在3.15~64.00μg/mL范围内、大黄素甲醚在1.70~33.92μg/mL范围内,其峰面积与浓度呈良好线性关系,5个成分的精密度RSD均小于2%,5个成分的平均回收率为102.1%,RSD均小于5%;大黄与黄连或黄芩配伍时可导致蒽醌类成分含量变化明显。结论:本研究建立的HPLC方法准确,重现性好,可用于大黄黄连泻心汤配伍研究时大黄蒽醌类成分的定量分析;大黄与黄连或黄芩配伍时蒽醌类成分变化明显,有无配伍黄芩对蒽醌类成分含量有显著影响。This is a study to determine 5 main compounds of rhubarb in Rhubarb and Coptidis Xiexin decoctions, using an assay method, in an attempt to compare the concentration of anthraqninones in different compatibility infusions. We developed a HPLC method, using a phenyl column (250mm×4.6mm, 5μm) under an ambient temperature, and using gradient elution with mobile phase of methanol-0.01% phosphoric acid and detected at 254 nm, to determine the concentration of 5 compounds in Rhubarb. The calibration curve of aloe-emodin, rhein, emodin, and chrysophanol indicating a good linearity was obtained within the range of 3.15-64.00μg/ml , and 1.70-33.92μg/ml of physcion. The RSD of 5 compounds were less than 2%. The average extraction recovery of the 5 main compounds was 102.1%, with an RSD less than 5%.The compatibility of rhubarb and coptis (scutellaris) has a significant effect on the concentration of anthraquinones. It is concluded that this method has been proved to be accurate and reliable for determining the concentration of anthraquinones in prescriptions containing Radix et rhizoma rhei. The assay results show that an increased or reduced scutellaris concentration in Rhubarb and Coptis Xiexin Decoction has a significant effect on the concentration of anthraquinones.
