摘 要:目的:探讨经IFN-γ刺激后,人外周血单个核细胞表达CD40的变化,以及经辛伐他汀干预后不同基因型单个核细胞表达CD40的影响。方法:随机选择42例健康志愿者,密度梯度离心法分离外周血单个核细胞并原代培养,用100μg/LIFN-γ及20μmol/L辛伐他汀对单个核细胞进行干预,流式细胞术检测单个核细胞培养24h后表达CD40的水平。聚合酶链反应限制性片段长度多态性(PCR-RFLP)检测这42位志愿者CD40-1C/T基因多态性,分析不同基因型个体外周血单个核细胞表达CD40的水平。结果:基础状态下单个核细胞培养24h后,CD40表达水平升高,且CC、CT、TT基因型单个核细胞CD40表达水平无显著差异(P>0.05);IFN-γ刺激状态下,单个核细胞CD40表达水平显著升高,且CC基因型升高的幅度显著高于CT、TT基因型(P<0.05);辛伐他汀和IFN-γ共同作用下,辛伐他汀可显著抑制IFN-γ诱导的CD40表达,但对CC、CT、TT基因型单个核细胞的抑制率没有统计学意义(P>0.05)。结论:CD40-1C/T多态性可以影响单个核细胞CD40表达水平,辛伐他汀可显著抑制IFN-γ刺激后CD40的表达,但对不同基因型单个核细胞的保护作用无显著性差异。Objective To explore the change of CD40 expression in peripheral blood mononuclear cells (PBMNC) with induction of IFN-γ, and the effects of simvastatin on expression of CD40 induced with IFN-γ in PBMNCs with different genotypes. Mothods Forty-two heahhy volunteers were enrolled, PBMNCs were separated from heparinized venous blood by density gradient centrifugation and then cultured with 100 μg/L IFN-γ or 20 μmol/L simvastatin for 24 h. The CD40 expression level was determined by flow cytometry. Genotypes of the CD40-1C/T were assayed by polymerase chain reaction-restricted fragment length polymorphism(PCR-RFLP). The expression levels of CD40 in PBMNC with different genotypes were analyzed. Results The levels of CD40 expression without intervention was increased after 24 h, however, there was no significant difference among CC, CT and TT genotypes (P 〉 0.05). The levels of CD40 expression in PBMNC in all genotypes were increased significantly after induction with IFN-γ and culture for 24 h, and the increase extent of CC genotype was significant higher than that of the other genotypes( P 〈 0. 05 ). Simvastatin could significantly inhibited the expression of CD40 in PBMNC induced with IFN-γ, while the inhibitory effects of Simvastatin had no significant difference among different genotypes. Conclusion CD40-1C/T polymorphism could influence the CD40 expression of PBMNC, Simvastatin could markedly inhibit the expression of CD40 in PBMNCs induced with IFN-γ, but there was no significant difference in the protective effects of Simvastatin among different genotypes of PBMNCs.
