摘 要:目的脂质过氧化对Wistar大鼠一氧化氮(NO)活性的影响。方法超氧阴离子、NO相关因素灌流离体大鼠主动脉,观察血管环一磷酸鸟苷(cGMP)水平和MDA含量的动态变化。结果与对照组比较,低浓度、高浓度H2O2灌流主动脉分别使血管条cGMP含量增加23.08%(P<0.05)、降低75.21%(P<0.01);预先应用NO合成底物L-Arg孵育,可以显著抑制高浓度H2O2灌流主动脉引起的cGMP水平下降的效应,使MDA含量降低;与较高浓度H2O2灌流组比较,L-NNA灌流使血管cGMP明显降低(P<0.05),MDA含量增加(P<0.05);NPS、SOD灌流可以拮抗高浓度H2O2抑制cGMP活性(P<0.01)的效应;并显著降低MDA含量(P<0.01)。结论低浓度和高浓度的脂质过氧化,分别促进和抑制血管NO的合成与释放,表明NO在一定范围内可以抑制脂质过氧化,进而对缺血、缺氧组织有明显的保护作用;同时超氧阴离子过剩与NO生成和功能缺陷有直接的相关性。Objective To probe into active effect on nitric oxide and peroxynitrite interacts with lipids in Wistar rat. Methods Wister rat's aorta in vitro is perfused by superoxide anion and NO relative factors and the change of vascular cGMP and MDA level was observed. Results Wister rat's aorta in vivo perfused by lower and higher concentration H2O2 , respectively made the vascular eGMP content increases 23.08% (P〈0.05) and decrease 75.21% (P 〈 0.01 ) as compared with control group. Wister rat' s aorta in vivo foreordain incubated by L-Arg, inhibited the decreased effect of cGMP level caused by higher concentration H2O2 perfusion and makes MDA content decrease. As compared with higher concentration H2O2 perfusion group, the vascular cGMP content decreases (P〈0.05) and M DA content increases (P〈0.05) in L-NNA group NPS and SOD perfusion can antagonise effect caused by higher concentration Ha 02 perfusion,make cGMP content increase (P〈0.01) and MDA content decrease (P〈0.01). Conclusion Wister rat's aorta in vitro perfused by lower and higher peroxynitrite interacts with lipids can promote and inhibit the production and release of NO respectively,which indicates NO may inhibit scavenging of superoxide and protect from ischemia and anoxia tissue;and simultaneity there is a direct relativity in vascular superoxide anion overmuch and NO production defect.
