参枸胶囊对小鼠学习和记忆功能及脂质过氧化损伤影响的研究

摘  要:目的:观察参枸胶囊对东莨菪碱、环己亚胺及乙醇所致的小鼠学习记忆获得、巩固和再现障碍及乙醇所致小鼠脂质过氧化损伤的影响。方法:使用跳台法观察小鼠的学习记忆功能、用TBA法测定MDA含量、NBT法测定SOD活性。连续灌胃给予参枸胶囊10 d,于实验的第9天训练,第10天测试,记录各动物3 min内的错误次数;MDA含量和SOD活性于末次给药16 h后断头取脑组织匀浆测定。结果:与生理盐水组比较,东莨菪碱、环己亚胺及乙醇模型组小鼠3 min跳台错误次数明显增加、乙醇模型组脑组织MDA含量显著升高、SOD活性显著下降;参枸胶囊8.4 g/kg组及脑复康组错误次数明显减少;脑组织MDA含量显著下降、SOD活性显著升高,其中参枸胶囊高剂量组尤为显著。结论:参枸胶囊对东莨菪碱、环己亚胺及乙醇所致的小鼠学习记忆获得、巩固和再现障碍及乙醇所致小鼠脂质过氧化损伤均有明显的改善作用。Objective: To observe the influences of Shengou Capsule on disorder of rat's study and memory function caused by scopolamine, cycloheximide; and lipid peroxidation damage caused by ethanol. Methods: Jumping stand method was used to observed rat's memory function, TBA method determined the content of MDA, NBT method determined the activity of SOD. intragastric administration for ten days successively, training on 9th day, testing on 10th, all wrong times were recorded in three minutes, the content of MDA and activity of SOD, from brain tissue, were determined after 16 hours last administration. Results: Compared with normal sodium, the 3 minutes wrong times of jumping stand increased obviously scopolamine, cycloheximide, ethanol model groups, the contents of MDA increased in ethanol model group and the activity of SOD reduced obviously; all these were opposite in Shengou Capsule groups(8 or 4 g/kg), especially obvious in high dosage group. Conclusion: Shengou Capsule has obvious improving effect on rat's study and memory disorder and lipid peroxidation damage.
