精脒促进E.coli非特异性转录的研究

摘  要:本文以EcoRI酶切的质粒pBR322为模板,在体外转录系统中研究了精脒对E.coliRNA聚合酶非特异性转录活性的作用。结果表明:0.50—10.00mM精脒对转录有明显促进作用,2.50mM作用最强;精脒的对数浓度与转录产物相对放射活性间呈正态分布曲线。In this paper,the effect of spermidine on the nonspecific transcription activity of E.coli RNA polymerase was studied.We used EcoRI-digested plasmid pBR322 as the template in vitro.The results demonstrated that transcription was markedly stimulated by 0.50-10.00 mM spermidine among which 2.50 mM spermidine had the strongest effect,and the relation of logarithm concentrations of spermidine with the relative radioactivity of the transcriptions was normal distribution curve.The study suggested that the effect mechanism of spermidine on transcription might be dualistic or even be multidimensional.
