体外培养小鼠骨髓巨核祖细胞的增殖和分化

摘　　要：以伴刀豆凝集素Ａ刺激小鼠脾细胞的条件培养基作为巨核细胞集落刺激因子（ＣＳＦ－ＭＫ）的来源，研究小鼠骨髓巨核祖细胞在体外培养中的增殖和分化。骨髓细胞经９ｄ培养结果表明，巨核细胞的数量在培养第３ｄ开始增高，５～７ｄ增殖达到高峰，之后逐渐下降；在培养的同一时间内，还可观察到分化程度不同的巨核细胞，提示巨核祖细胞是具有年龄结构的细胞群及ＣＦＵ－Ｍｅｇ细胞池具有不均一性的特征。The megakaryocyte has fastidious growth requirement in vitro.ConA-stimulated spleen cell conditioned medium has been used to supply the colony stimulating factor megakaryocyte (CSF-MK) required for culture so as to observe the growth and development of megakaryocyte.C57BL/6J mouse bone marrow cells were incubated in culture for up to 9 days.The results showed that the number of megakaryocytes increased at day 3,to its peak between day 5 and 7 and progressively decreased subsequently.Megakaryocyte in different mature stages were observed morphologically in same time of culture.It is suggested that the CFU-Meg was a group of aged structure cells and the pool of CFU-Meg showed the character of uniform.
