摘 要:目的观察消可宁颗粒剂对高糖条件下培养的人脐静脉血管内皮细胞(ECV-304)的保护作用。方法苏木精-伊红染色观察体外培养的不同消可宁浓度下的ECV-304细胞形态学改变和四甲基偶氮唑盐法(MTT)测定ECV-304细胞增殖活性。结果①高糖对照组ECV-304细胞生长抑制,变小变圆,出现不同程度的空泡变性。各消可宁组ECV-304细胞出现不同程度的细胞恢复。②80 g/L和100 g/L消可宁组可减轻高糖对ECV-304细胞的生长抑制,与高糖对照组比较具有显著性差异(P<0.05)。结论高糖引起ECV-304细胞损伤,对其生长增殖具有明显的抑制作用。消可宁可有效减轻高糖对ECV-304细胞的损伤和生长抑制,从而保护内皮细胞。Objective To investigate the protective effect of Xiaokening granules on umbilical vein endothelial cells (ECV- 304) induced by high glucose. Methods ECV -304 cell were cultured in vitro at different concentrations of Xiaokening and high glucose. (1)Observations of the ECV -304 cells stained with hematoxylin and eosin (HE) was performed for morphological changes. (2)The proliferation activity of ECV - 304 cells cultured in vitro at different concentrations was determined by using MTT. Results (1)It was found that ECV - 304 ceils in high glucose control group were inhibited in growth and became small and round, with varied vacuolar degenerations. In each of Xiaokening groups, the ECV - 304 cells exhibited recovery to certain extent. (1)In Xiaokening groups, in contrast to high glucose group, inhibitory effect on the proliferation of ECV were reduced, especially in 80 g/L and 100 g/L groups (48 hours groups) and the differences were considered statisticaUy significant ( P 〈 0.05). Conclusion High glucose causes injury to ECV - 304 cells and remarkably suppresses their proliferation, while Xiaokening can effectively reduce the cellular injury and proliferation inhibition resulting from high glucose to ECV -304, hence rendering protective effects on endothelial cells.
