摘 要:目的研究脱氢表雄酮(DHEA)对BXSB狼疮性小鼠脾脏淋巴细胞凋亡的影响,并探讨其作用机理。方法运用流式细胞术、免疫双荧光染色法检测脾脏淋巴细胞凋亡。结果当DHEA的浓度为10^-8moL/L时,淋巴细胞凋亡率降低,与对照组比较有差异有统计学意义(P〈0.05),细胞坏死率差异无统计学意义(P〉0.05);DHEA与西药组或中药组联合应用对淋巴细胞凋亡抑制作用更加显著,差异有统计学意义(P〈0.01)。结论脱氢表雄酮可明显抑制BXSB小鼠脾脏淋巴细胞凋亡率。Objective To study the effect of dehydroepiandrosterone(DHEA) on apoptosis in spleen lymphocytes of BXSB mice with Systemic lupus erythematosus (SLE) and analyze the mechanism of treatment. Methods The apoptosis of spleen lymphocytes of BXSB mice treated with DHEA was examined by the flow eytometric analysis and immunofluoreseence double-staining method. Results Treated with 10^-8mol/L DHEA,the apoptotic rates of spleen lymphoeytes in DHEA treated group were significantly lower from that in the control group (P 〈0.05). there was no significantly difference in cellular necrosis between these groups(P 〉 0. 05 ) ; When traditional chinese medicine group and western medicine group were combinated with DHEA , there were significantly different( P 〈 0.01 ). Conclusion DHEA can restrain the excessive apoptosis of spleen lymphoeytes in BXSB mice with SLE.
