摘 要:目的研究血管活性肠肽(VIP)对大鼠腹腔肥大细胞功能的影响。方法分离、纯化SD大鼠腹腔肥大细胞,应用不同浓度的VIP、VIP受体抑制剂(L-8-K)和VIP类似物(VIP6-28)作用于大鼠腹腔肥大细胞后,同位素放射液态闪烁法检测肥大细胞的组胺释放、45Ca内流和细胞内cAMP含量的变化。结果5×106mol/L的VIP、VIP6-28作用后大鼠腹腔肥大细胞组胺释放及45Ca内流明显增加、细胞内cAMP水平明显降低,并且这种变化与VIP、VIP6-28之间呈剂量效应关系;L-8-K对VIP诱导的肥大细胞组胺释放、45Ca内流的变化无明显影响。结论VIP通过非受体依赖途径引起肥大细胞钙内流增加、细胞内cAMP水平下降,进一步诱导肥大细胞脱颗粒、释放组胺等生物活性物质,产生生物学效应。Objective To investigate the role of of VIP and VIP analog in the physiological function of rat peritoneal mast cells. Methods The SD rat peritoneal mast cells were isolated, purified and dispensed. Isotope liquid scintillation counting was used to measure the content of histamine release, ^45Ca influx and cAMP level of the rat peritoneal mast cells, induced by different concentration of VIP, L-8-K and VIP 6-28. Results After the rat peritoneal mast cells were treated with 5 × 10^-6 mol/L VIP or VIP 6-28 ,the histamine release and ^45Ca influx of the mast cells were increased, and the cAMP in the mast cells was decreased, remarkably;and these changes were dose-dependent with VIP and VIP 6-28. There were no effects of L-8-K on the VIP-induced changes of histamine release, and 45Ca influx of the rat peritoneal mast cells. Conclusion VIP induces degranulation and histamine release from rat peritoneal mast cells by the receptorindependent calcium influx increase and intraeellular cAMP decrease pathway.
