摘  要:目的探讨白芍总苷(TGP)对糖尿病大鼠肾组织中p38MAPK磷酸化及NF-κBp65表达的影响。方法采用链脲佐菌素(STZ)诱导大鼠糖尿病模型。随机分为对照组、模型组、TGP组(50、100、200mg/(kg.d)),8周后应用West-ern免疫印迹方法检测肾组织p38MAPK磷酸化(p-p38)及NF-κBp65表达。结果TGP给药(50、100与200mg/kg)8周可明显降低大鼠尿白蛋白排泄。TGP给药(50mg/kg)大鼠肾小球平均容量(VG)明显低于模型组,肾小管一间质损伤指数(TⅡ)较模型组有所下降,但差异无显著性;TGP给药(100、200mg/kg)VG与TⅡ均明显低于模型组。Western印迹显示模型组肾组织p38MAPK磷酸化及NF-κBp65表达显著增强,TGP给药(50、100与200mg/kg)8周肾组织p38MAPK磷酸化表达下降30.8%、24.0%、60.1%,NF-κBp65表达下降27.8%、45.9%、59.0%。结论TGP可明显改善糖尿病大鼠早期肾损害,其机制可能与部分抑制糖尿病大鼠肾组织p38MAPK和NF-κB激活有关。Objective To investigate the effect of Total glucosides of paeony(TGP)on activation of p38 mitogen-activated protein kinase(p38 MAPK)and nuclear factor-κB p65(NF-κB p65)in the kidney from experimental diabetic rat.Methods Diabetes was induced with streptozotocin in rats,and TGP(50,100,200 mg/(kg·d))was orally administered once a day for 8 weeks.The expression of phosphorylation of p38(p-p38)MAPK and NF-κB p65 were determined by western blot analysis in the kidney.Results Elevated 24 h urinary albumin excretion rate was markedly attenuated by TGP treatment with 50,100 and 200 mg/kg.Elevated glomerular volume was significantly attenuated by TGP treatment with 50,100 and 200 mg/kg,and increased indices for tubulointerstitial injury were only ameliorated by TGP treatment with 100 and 200 mg/kg.Densitometric analysis of the Western blot showed increase in the amount of p-p38 and NF-κB p65 in the kidney from diabetic rats with respect to normal rats,TGP treatment with 50,100 and 200 mg/kg could reduced p-p38 and NF-κB p65 expression by 30.8%,24.0%,60.1% and 27.8%,45.9%,59.0%,respectively.Conclusion Our data suggest that TGP treatment ameliorates early renal injury via the inhibition of the overexpression of p-p38 and NF-κB p65 protein in the kidney from diabetic rats.
