摘  要:目的:探讨脂肪源性干细胞作为骨组织工程的种子细胞在修复兔颅骨缺损中的应用前景.方法:提取兔脂肪源性干细胞,成骨诱导并检测,将成骨诱导前后的脂肪源性干细胞种植到自制松质骨支架上,将细胞-支架复合物和单纯支架材料分别移植至兔颅骨缺损部位.结果:与未诱导的脂肪源性干细胞复合支架、单纯支架以及空白处理对比,成骨诱导后的脂肪源性干细胞复合支架修复骨缺损面积最大,差异具有统计学意义(P<0.05).结论:脂肪源性干细胞可作为骨组织工程的种子细胞,其成骨诱导后与自制松质骨复合植入体内,能显著促进骨缺损的修复.AIM: To investigate the use prospect of adiposederived stem cells(ADSCs) as seed cells in tissue-engineering for the repair of rabbit skull defect. METHODS: ADSCs were collected and induced to differentiate along osteogenic lineages. The osteogenic-induced and non-induced ADSCs were seeded respectively onto self-made heterogeneity cancellous bone scaffold. The cell-scaffold compounds and simple scaffold were transplanted to skull defect of rabbits. RESULTS: Compared with non-induced ADSCs-scaffold, simple scaffold and vacant control groups, there was significantly greater healing in skull defects in the osteogenicinduced ADSCs group. CONCLUSION: ADSCs can be used as seed cells in bone-tissue-engineering. The osteogenic-induced ADSCs compounded with self-made heterogeneity cancellous bone scaffold can accelerate the repair of bone defect significantly.
