摘 要:目的建立一种简便的一室模型药物血管外给药的药动学参数计算方法;方法间隔τ时间前后给予相同的剂量d,设计4个取样点(t=0、τ/2、τ和τ+τ/2),根据给定的药动学参数(ka、k和V/F)计算各取样点的理论血药浓度(c0、c1、c2和c3),推算ka、k和V/F的近似值,并分析各参数的相对误差百分率(E%),计算E%<5%的τ值;结果参数推算值与给定值基本一致,ka、k值虽对E%有一定的影响,但在设置τ的最小取值后可以确保V/F、ka和k的E%小于5%。结论双剂量四点取样法可以用于获取一室模型血管外给药的药动学参数。OBJECTIVE To obtain the pharmacokinetic parameters of one-compartment model drug by extra-venous administration.METHODS The double-dose and four-sampling method was used to calculate approximate values of parameters(ka,k and V/F)by theoretic plasma concentration(c0,c1,c2 and c3) of four sampling points(t=0,τ/2,τ and τ+τ/2) on provided pharmacokinetic parameters,and relative error percentage(E%)of the parameters were analyzed,the interval time(τ) of E%〈5 was decided.RESULTS The calculated value of parameters basicall was in accordance with provided.Although E% was influenced from the parameters ka and k,but E%〈5 was confident when minimum value of τ was set.CONCLUSION The double-dose and four-point method can be used to obtain the pharmacokinetic parameters of one-compartment model drug by extra-venous administration.
