摘 要:目的观察左归复方(滋阴益气活血解毒组方)干预治疗MKR小鼠后糖代谢、血清胰岛素及胰岛B细胞胰岛素表达的变化。方法40只MKR小鼠经鉴定后,随机分为模型组,左归复方高、低剂量组,阳性对照组(文迪雅),每组10只,另设10只C57小鼠为野生鼠组。各药物组分别以相应剂量连续给药30d,模型组和野生鼠组灌胃蒸馏水。于给药前、第15日及第30日分别作空腹血糖测定;末次给药后0.5h心脏取血,放射免疫法测血清胰岛素,免疫组织化学SABC法检测胰岛B细胞胰岛素表达。结果左归复方干预治疗后,其高剂量具有显著的降低MKR小鼠空腹血糖、改善高胰岛素血症的作用。MKR小鼠胰岛B细胞胰岛素表达显著高于C57野生型小鼠;左归复方高剂量组干预治疗后胰岛B细胞胰岛素表达显著下调;左归复方高剂量组下调胰岛B细胞胰岛素表达与对照药文迪雅作用相当。结论左归复方具有改善MKR小鼠糖代谢、减轻胰岛B细胞的工作负荷、减少胰岛素表达及分泌的作用。Objective: To observe the expression and secretion of insulin in the pancreas islet B cells of MKR mice treated by Zuogui Recipe. Methods: 40 MKR mice were randomly devided into model group(MG), low Zuogui Recipe group(LZG), high Zuogui Recipe group(HZG) and positive control group(PCG). LZG, HZG and PCG were treated by corresponding drug for 30d. Using C57 mice as control, the fasting blood glucose level and the serum insulin level were determined, and SABC immunohistochemical study was on the expression of insulin. Results: After treated with Zuogui Recipe, the MKR mice were significantly ameliorate hyperglycemia, decreased serum insulin. The expression of insulin was stronger in the pancreas islet B cells of MKR mice than C57 mice. After treated with of Zuogui Recipe, the expression of insulin was significantly decreased in the HZG. And there was no significantly difference between HZG and PCG. Conclusion: Zuogui Recipe can decrease hyperglycemia, and decrease the working pression of pancreas islet B cells through decreasiong the expression and secreation of insulin.
