摘 要:目的:分析阿霉素肾病大鼠肾组织结缔组织生长因子(connective tissue growth factor,CTGF)蛋白及其mRNA的变化,研究三七总皂苷对其干预作用及机制。方法:采用单侧肾切除阿霉素肾病模型。将45只大鼠随机分为5组,正常组、假手术组、模型组、苯那普利组和三七总皂苷治疗组。术后10周处死大鼠,观察肾组织病理改变,应用免疫组织化学方法和原位杂交法检测肾组织CTGF及其mRNA的表达。结果:正常组及假手术组肾小球系膜细胞、肾小管上皮细胞以及肾间质细胞胞浆内有少量CTGF蛋白及其mRNA的黄色颗粒状阳性反应物,模型组上述部位的CTGF蛋白及其mRNA呈强阳性表达,三七总皂苷组上述指标表达较模型组显著减少,有统计学差异(P<0.05),与阳性药物组比较差异不显著。结论:三七总皂苷可能通过抑制CTGF蛋白及其mRNA的过度表达而减缓肾纤维化病程进展。Objective :To investigate the change of connective tissue factor (CTGF) in adriamycin nephropathy rats, and study the effect of Panaxnotoginseng on adrimysin - induced nephropathy rats and its possible mechanism. Methods : One side nephrectomize and ADR was used. 45 Wistar rats were randomly divided into 5 groups : normal control group, adriamyein nephropathy group, sham operation group, Benazepril treated group and Panaxnotoginseng treated group. Renal tissue were examined by microscopy at the 10th week after operation. Immunohistochemistry and in - situ hybridization was used to measure the expression of CTGF protein and CTGF mRNA in the nephridial tissue. Results : The expression of CTGF protein and CTGFmRNA in the nomal and sham operation group is low in the intracytoplasm of Mesangial cells,renaltubular epithelial cells and renal interstitial cells. Compare with sham operation group, the expression of CTGF protein and CTGFmRNA as yellow particulate in the mode group was markedly increased, and the fibrosis of nephridial tissue is obvious. Panaxnotoginseng could significantly decrease the expression of CTGF protein and CTGFmRNA ( P 〈 0.05), and there was no significant difference between Panaxnotoginseng treatment group and Benazepril treatment group. Conclusions : Panaxnotoginseng may suppress the development of fibrosis, at least in part via down - regulating the expression of protein and mRNA of CTGF.
