摘  要:目的:考察河北不同产地知母药材中芒果苷及菝葜皂苷元的含量。方法:分别采用HPLC法和HPLC-ELSD法测定芒果苷和菝葜皂苷元的含量。测定芒果苷以水∶乙腈∶0.85%磷酸(100∶25∶5)为流动相;流速为0.6mL/min;柱温30℃;检测波长为258nm。测定菝葜皂苷元以甲醇∶水(95∶5)为流动相;流速为1.0mL/min;柱温30℃;用蒸发光散色检测器检测,漂移管温度85℃。结果:河北不同产地知母药材中菝葜皂苷元含量为1.51%~1.73%,芒果苷含量为0.11%~0.73%。结论:本法测定知母药材中菝葜皂苷元及芒果苷含量,方法简单、快速、准确、重现性好,方法学考察符合含量测定的要求,可以用来控制知母药材质量。Objective:To establish the determination of mangiferin and sarsasapogenine in Rhizoma Anemarrhenae from different areas. Methods: Determination of mangiferin and sarsasapogenine in Rhizoma Anemarrhenae were by HPLC and HPLC - ELSD. The mobile phase were water - aeetonitrile - 0.85% phosphoric acid( 100: 25:5 ) and methanol - water( 95: 5 ) ,with the flow rate were 0.6mL/min and 1.0mL/min, at 30℃. The detection wavelength of mangiferin was at 258nm. An evaporative light - scattering detector (ELSD) was used as detector to determine sarsasapogenine with the drift tube, temperature of 85℃. Result:The content of mangiferln and sarsasapogenine in Rhizoma Anemarrhenae from different areas were 0.11% - 0.73% and 1.51% - 1.73%. Conclusion:The method can be used for the equality control of Rhizoma Anemarrhenae from different areas,the restdt have provide a scientific basis for processing Rhizoma Anemarrhenae.
