摘 要:目的:通过比较和分析中药药对黄芩柴胡的合煎、分煎以及配方免煎颗粒剂的HPLC紫外图谱信息,探求不同调配方法对药物组分的溶出度及组分改变的影响。方法:逆相色谱柱:RPC18(Cadenza CD-C18# ZB0703C4.6mm×150mm,4μm)。流动相:A=0.1%TFAH2O,B=0.1%TFAH2O/CAN=1/9。梯度:A/B=100/0~90/10in5minA/B=90/10~60/40innext55min。流速:1.0mL/min;检测波长:215nm;进样量20μL。结果:各组合煎时,药对中特定组分的溶出度增加,部分主要组分高于分煎组2倍以上。免煎颗粒的溶出度与分煎接近。各组合煎与分煎或颗粒剂之间相比,出现了少量低丰度峰的增减改变。结论:合煎能使药物的成分溶出度增加,但组分改变不显著。免煎颗粒有推广的可能性,但在量的等效性方面。还有必要加以进一步考察。Objective :Through the ultraviolet spectrum of the mixed decoction (mix the traditional Chinese herbs together, and then boil them in water), the separated decoction ( boil the single traditional Chinese herb in water separately, and then mix them together), and extracted granule of couple of Redix Scutellariae and Redix Bupleuri, we analysis the overall ingredients, compare the differences,in order to investigate the influence of different decoct methods, on component and solute quantity. Methods : Column : RPC18 4.6mm × 150mm ( Cadenza CD - C18 #ZB0703 C). Dilute every decoction with A (0.1% TFA H2O) HPLC. method:Solvent of Mobile Phase:A =0.1% TFA H20,B =0.1% TFA H20/CAN = 1/ 9. Gradient :A/B = 100/0- 90/10 in 5min A/B = 90/10 - 60/40 in next 55min. Flow rate was 1. OmL/min and the detection wavelength was 215 nm. Results : The dissolution rate of mixed decoctions is larger than others in total and some special ingredients also increased. The dissolution rate of herb extract is close to that of separated decoctions. Conclusion: Mixed decoction can make the dissolved matter of herbs medicine increase partially, but the change of components is not obviously. Despite extracted granule has the value to be popularized,there also being a necessity to conduct a further investigation in the equivalence of quantity. The ingredients and the dissolution rate of the ingredients of the mixed decoctions can be different from that of the separated decoctions. While, the difference of dissolution rate between herbs extracts and separated decoctions is not notice.
