摘  要:目的:研究三七皂甙对大鼠慢性缺氧性肺动脉高压的作用。方法:采用间断常压缺氧法制备大鼠慢性缺氧性肺动脉高压模型,腹腔注射三七皂甙(25,50和100mg·kg-1·d-1),连续给药21天。结果:三七皂甙能产生剂量依赖性地抑制慢性缺氧性肺动脉高压的作用,其抑制率分别为15.9%、43.9%和73.4%,并能明显抑制右心室肥厚,其抑制率分别为22.1%、55.6%和83.3%,而对颈动脉血压无明显的影响。三七皂甙(200g·L-1)能明显抑制血小板生长因子所诱导的离体肺动脉平滑肌细胞DNA的合成。结论:三七皂甙有抑制慢性缺氧性肺动脉高压的作用,可能是通过抑制血小板生长因子促肺动脉平滑肌细胞增殖而抑制肺血管结构重建来实现的。AIM:Effects of panax notoginseng saponins (PNS) on chronic hypoxic pulmonary hypertension in rats was investigated. METHODS: The model of rat chronic hypoxic pulmonary hypertension was made by method of intermittent isobaric hypoxia. The rats were injected intraperitoneally with (PNS 25 mg·kg -1 ·day -1 ,50 mg·kg -1 ·day -1 and 100 mg·kg -1 ·day -1 ) for 21 days. RESULTS: The PNS could inhibite develepment of chronic hypoxic pulmonary hypertension in dose dependent manner. The inhibitory rates of PNS on increase of pulmonary artery pressure induced by chronic hypoxia were 15 9%, 43 9% and 73 4% and on hypertrophy of right ventricle induced by chronic hypoxia were 22 1%, 55 6% and 83 3% respectively without effect on cervical artery pressure. The synthesis of DNA in pulmonary artery smooth muscle cells induced by platelet derived growth factor (PDGF) was decreased by PNS(200 g·L -1 ). CONCLUSIONS: These results suggested that PNS could inhibite chronic hypoxic pulmonary hypertension through decreasing proliferation of pulmonary artery smooth muscle cells and remodelling of pulmonary vessels.
