摘  要:目的:观察硝苯地平对自发性高血压大鼠(SHR)的动脉血管平滑肌细胞(VSMCs)增殖、c-fos原癌基因表达和细胞内肌醇磷脂含量的影响,以进一步阐明硝苯地平的抗高血压作用机制。方法:以MTT法、斑点印迹杂交和细胞内肌醇磷脂含量测定方法与对照组比较硝苯地平对细胞的影响。结果:硝苯地平是VSMCs异常增殖的有效抑制剂,并能明显阻止SHR的VSMCs内c-fos过度表达;研究也显示,硝苯地平显著地减小细胞内肌醇磷脂的过量增加。结论:硝苯地平能抑制SHR的VSMCs的c-fos过度表达和细胞异常增殖,而这一作用可能与其阻止细胞内肌醇磷脂的过量增加有关。AIM: To further elucidate the antihypertensive mechanism of nifedipine in SHR. METHODS: The effect of nifedipine on production of inositol phosphates, c fos expression and proliferation of vascular smooth muscle cells (VSMCs) from SHR were investigated in this study. RESULTS: It was shown that nifedipine was a strong inhibitor of SHR VSMCs growth, and nifedipine suppressed serum induced expression of c fos protooncogene mRNA in VSMCs from SHR. Additionally, nifedipine could signicantly block serum stimulated production of inositol phosphates in VSMCs from SHR. CONCLUSIONS: It is suggested that nifedipine can inhibit VSMCs proliferation and induction of c fos mRNA in SHR VSMCs stimulated by serum, which might be related to a significant decrease in inositol phosphate formation in the presence of nifedipine.
