摘  要:目的观察经冠脉联合导入骨髓间充质干细胞(MSCs)及碱性成纤维细胞生长因子(bFGF)基因对急性心梗心功能的影响。方法开胸结扎法制作小猪急性心肌梗塞模型(11只),1周后行冠状动脉造影,随机分为对照组和实验组,分别经冠脉导入生理盐水10ml,含1×107MSCs的生理盐水10ml和pcDNA3-bFGF2000ug。于心梗前、心梗后1周以及心梗后5周行心脏超声检查,心梗后5周再次测定左室舒张末压。结果所有动物均成功建立急性心肌梗死模型,心梗后1周左室射血分数明显下降,5周后实验组心功能较对照组改善。结论联合导入骨髓间充质干细胞及碱性成纤维细胞生长因子基因可改善急性心肌梗死的心功能。Objective To observe the cardiac function influenced by combined bone marrow mesenchymal stem cells and basic fibroblast growth factor therapy in swines with myocardial infarction. Methods To establish animal models for acute myocardial infarction (AMI) by using LAD ligation(11 animals). Experimental animals were devided into two groups: control group and experimental group. Coronary angiography was performed one week after myocardial infarction. 10ml normal saline, 1 x 107 MSCs and 2000ug bFGF-expressing MSCs were infused into myocardium by selective coronary angiography. Left ventricular ejection fraction(LVEF) were measured before AMI, and 1 week and 5 week after AMI. Left ventricular diastolic end pressure measured 5 week after AMI. Results All animals occurred with acute myocardial infarction. LVEF decreased after AMI. Cardiac function improved in experimental group 5 week after MI. Conclusion Combined bone marrow mesenchymal stem cells and basic fibroblast growth factor therapy can improve cardiac function in myocardial infarction.
