摘 要:目的探讨1-磷脂酰肌醇3-激酶-丝氨酸-苏氨酸蛋白激酶B(PI3K-AKT)信号传导通路效应物在胎鼠肺发育过程中的表达。方法运用Western蛋白印迹方法检测不同胎儿期鼠肺上皮细胞中AKT、磷酸化AKT(pAKT)及相关蛋白的表达。运用免疫组织化学方法检测不同胎儿期鼠肺上皮细胞中pAKT的表达水平。结果Western蛋白印迹法显示,AKT在不同胎儿期肺上皮细胞中的蛋白表达无明显差异;第12天胎鼠肺pAKT高度表达,第14天开始减少;第12天胎鼠肺cyclinD1高度表达,第14天开始减少;第12天胎鼠肺SPC低度表达,第14天开始逐渐增加。免疫组织化学法显示,第14天胎鼠肺上皮细胞中pAKT高度表达,第18天胎鼠肺上皮细胞中pAKT无明显表达。结论PI3K-AKT信号传导通路通过许多生物过程在胎鼠肺发育中起重要作用。Objective To study the effect of PI3K-AKT signal pathway in fetal mouse lung development. Methods The expression of AKT, phosphorylated AKT (pAKT) and the downstream proteins were examined with Western blotting and immunohistochemistry in the epithelium of fetal mouse lung at different time points. Results Western blotting showed that the expression of AKT was not significantly changed in embryonic lungs during development, while phosphorylated AKT (pAKT) was highly expressed on embryonic day 12 (E12) and started decreasing from embryonic day 14 (E14). CyclinD1 was highly expressed on E12 and started decreasing from E14. SPC was lowly expressed on E12 and started increasing from E14. Immunohistochemieal study showed that pAKT was expressed in the respiratory epithelium on E14. However, pAKT was not obvious expressed on E18. Conclusion The PI3K AKT signal pathway plays a pivotal role in mouse lung development through various biological processes.
