灯盏花素对凝血酶诱导内皮细胞基质金属蛋白酶-2表达的影响

摘　　要：目的探讨不同浓度的灯盏花素对凝血酶诱导的人脐静脉内皮细胞(HUVECs)基质金属蛋白酶-2(MMP-2)的表达及活性的影响。方法体外培养HUVECs,待细胞生长到融合状态时加不同浓度的灯盏花素(17.5、35.0、70.0μg/mL),1 h后加入凝血酶(4 kU/L),作用24 h后采用明胶酶谱法检测MMP-2的活性,逆转录聚合酶链反应(RT-PCR)技术测定MMP-2 mRNA的表达。结果凝血酶可诱导HUVECs MMP-2的表达;不同浓度的灯盏花素可抑制凝血酶诱导的MMP-2的表达(P<0.01)。结论灯盏花素可抑制凝血酶诱导的HUVECs MMP-2的表达;可能对抗动脉粥样硬化,稳定硬化斑块、防治斑块破裂和预防急性冠状动脉综合征的发生产生有益的作用。Objective To investigate the effects of Breviscapine at different concentrations on expression of matrix metalloproteinase-2 （MMP-2） induced by thrombin in eultured human umbilieal vein endothelial cells（HUVECs）. Methods The hreviscapine of different concentrations （17.5,35.0,70.0 μg/mL）were conincubated with cultured HUVECs for 24 hours after the HU- VECs were stimulated for 1 hour by thrombin（4 kU/L）. Expression of MMP-2 were determined by zymography and reverse transeription-polymerase chain reaction （RT-PCR）. Results Thrombin induced expression of HUVECs MMP-2. Breviscapine inhibited the expression of MMP-2 induced by thrombin （P〈0.01）. Conclusion Breviscapine could decrease the expression of MMP-2 in thrombin-induced HUVECs,which could be beneficial for the anti-atherosclerosis , atherosclerotie plaque stability, the prevention of plaque rupture and the occurrence of acute coronary syndrome.
