摘 要:目的建立同时测定五味消毒饮口服液中绿原酸和咖啡酸含量的方法。方法采用RP-HPLC法,Irregular-H C18(4.6mm×250mm,10μm)柱,流动相:乙腈-质量分数为O.04%的磷酸水溶液(体积比为12:88),流速:1.0mL·min^-1。,检测波长:327nm,柱温:25℃。结果绿原酸与咖啡酸分别在4.08~40.8mg·L^-1(r=0.9996)和0.1561.56mg·L^-1(r=0.9998)内线性关系良好,平均回收率分别为99.7%和99.3%,RSD分别为0.52%和0.27%(n=9)。结论所建立的方法可用于五味消毒饮口服液中绿原酸和咖啡酸的测定。Objective To establish an RP-HPLC method for simulaneous determination of chlorogenic acid and caffeic acid in Wuweixiaoduyin oral solution. Methods Irregular-H C18 column (4.6 mm × 250 mm, 10μm) was used with acetonitrile-0.04 % phosphoric acid solution( V: V = 12:88) as the mobile phase at a flow rate of 1.0 mL· min^-1. The detection wavelength was set at 327 nm and column temperature was 25 ℃. Results The calibration curve was linear in the range of 4.08 - 40.8 mg·L^- 1 ( r = 0. 999 6) and 0.156 - 1.56 mg·L^-1 ( r = 0.999 8) for chlorogenic acid and caffeic acid, respectively. The average recoveries were 99.7% and 99.3 % with RSD of 0.52 % and 0.27% ( n = 9), respectively. Conclusions The method is simple, accurate and reliable, which may be used for simultaneous determination of chlorogenic acid and caffeic acid in Wuweixiaoduyin oral solution.
