补肾中药改善骨质疏松大鼠骨骼强度的作用机制

摘　　要：目的:评价补肾中药改善去卵巢大鼠骨强度的作用效果和机理。方法:将48只6月龄健康雌性SD大鼠随机分为假切除组、模型组、补肾方组和雷罗昔芬组。大鼠灌胃给药3个月后处死,观察股骨骨矿密度、生物力学指标、骨组织形态计量学指标和骨超微结构指标等的变化情况。结果:①模型组骨密度为(0.250±0.04)g.cm-2,显著低于假切除组(0.302±0.03)g.cm-2,补肾方组为(0.268±8.34)g.cm-2。②模型组最大载荷(108.85±9.63)N,显著低于假切除组(121.07±8.34)N,补肾方组最大载荷为(122.91±8.72)N,与模型组比较有明显统计学差异。③补肾方能明显改善骨小梁面积、骨小梁厚度、骨小梁数目、骨小梁分离度和游离末端等形态计量学参数。④补肾方在一定程度上能保护骨结构。结论:补肾方能延缓骨结构的退变,有效改善去卵巢骨质疏松大鼠骨强度。Objective:To appraise the effect and mechanism of Chinese crude drug for invigorate the kidney to improve bone strength of rats with ovaries removed.Methods:48 cases of healthy and male SD rats with six months were randomly divided into four groups,group A (notho-remove group),group B (model group),group C (Chinese crude drug group) and group D (Raloxifene group).Drugs were given by intragastric admin istration for rats and they were killed three months later.Some changes on bone mineral density (BMD) and indexes of vitodynamics,tissue mor- phometry and fine structure in femoral bone were observed.Result:On BMD,those of group A,group B and group C were respectively (0.30 ### 0.03)g.cm-2,(0.250±0.04)g.cm-2and (0.268±8.34)g.cm-2,group B was obviously infra to group A.On max load,the three groups above were respectively (121.07±8.34) N,(108.85±9.63) N and (122.91±8.72) N,the same was seen on group A and group B.There had obvious statistical discrepancy on comparison of group B and group C.In group C,some morphometry parameters could be improved obvious ly,such as areas,thickness,numbers,resolving powers and liberating extrems of bone trabecula.It could protect bone architecture on some de gree.Conclusion:Chinese crude drug for invigorate the kidney could postpone cataplasia of bone architecture and improve bone strength of osteo porosis rats with ovaries removed.
