金樱子对IgA肾病大鼠肾脏组织蛋白质表达和肾脏功能的影响

摘　　要：目的:了解金樱子对IgA肾病肾脏功能及肾脏组织蛋白质表达的影响。方法:通过口服牛血清白蛋白(BSA)酸化水及尾静脉注射葡萄球菌肠毒素(SEB),建立IgA肾病大鼠模型,连续用药4周后,采用表面激光解吸离子化飞行时间质谱(SELDI-TOF-MS)技术扫描分析肾脏组织蛋白质的表达变化,并用生化自动仪测定血清白蛋白(Alb)、尿素氮(BUN)及肌酐(Cr)含量。结果:共扫描到70个蛋白峰,其中3个蛋白质在模型组中的表达有降低趋势,经过金樱子水醇萃取成分作用后表达增加;9个蛋白质在模型组中的表达有升高趋势,经过金樱子水醇萃取成分作用后荷质比为1033.16Da、9370.88Da的蛋白质表达降低;而分子量为7624.95Da、15229.41Da的蛋白质在治疗组中的表达高于模型组和正常组。Cr在三组中的含量分别为:正常组(24.34±2.11)μmol/l、模型组(33.61±3.01)μmol/l、治疗组(26.17±2.26)μmol/l。结论:金樱子能够调节IgA肾病大鼠肾脏组织蛋白质的表达,增加血清Cr的清除,改善肾脏功能。Objective： To study the effect of Rosa Laevigata Michx（RLM） on the protein expression and rats renal function of IgA nephropathy.Methods： 36 rats were divided into normal group,model group and treated group at random, IgA nephropathy model was set up by orally administration of BSA and intravenously injection of SEB, then treated with RLM. Protein from rats renal tissue was extracted, and scanned by SELDI-TOF-MS.The level of serum albumin,Bun and Cr was determined by automatic biochemical analyzer. Results：70 kinds of different molecular weigh （MW） protein was found.3 kinds of protein expression decreased in model group,and increased after treated with RLM; Other 9 kinds of protein enhanced, after treated,the expression of 1 033.16 Da,9 370.88 Da decreased ;the expression of protein with molecular weight of 7 624.95 Da,15 229.41 Da in treated group were higher than that in model group and normal group.The Cr content among the three groups was （24.34± 2.11）μmol/1,（33.61±3.01）μmol/1 and （26.17±2.26）μmol/1.Conclusion： The RLM can regulate the protein expression of renal tissues in IgA nephropathy rat, improve the Cr clearance and renal function.
