摘 要:目的:探讨递增运动负荷训练后大鼠心室肌蛋白质组的表达特征,为进一步筛选对运动性疲劳的研究具有重要意义的蛋白提供实验依据。方法:将10只SD雄性大鼠随机分为对照组和实验组。实验组经过7周的递增运动负荷训练(最后一次为力竭)后与对照组同时麻醉处死,取心室肌,提取心肌组织的全蛋白进行双向凝胶电泳分离。运用Bio-radPDquest图像分析软件对图谱进行分析。结果与结论:在电泳图谱上运动组可见蛋白质点338±17个,对照组可见352±17个。运动后差异表达的蛋白质点共有99个。运动后消失的点有5个,表达量上调4倍以上和下调幅度超过80%的点各12个,表达量上调1倍以上和下调幅度超过50%的点分别有52个和42个。递增负荷运动训练后,大鼠心室肌蛋白质组发生了明显的质变和量变。运动后表达量差异比较大的蛋白质点的分子量主要集中在35~57kD,等电点主要集中在6.0~7.4和8.6~9.6,在这个范围的蛋白质多为能量代谢酶,提示心室肌能量代谢的改变可能是运动引起心肌疲劳的原因。Objective To probe into the feature of rats' ventricular muscle proteome after incremental load training for screening out some proteins that are responsible to and meaningful in causing exercise-induced fatigue. Methods Ten SD rats was randomly divided into control and exercise group, the later underwent a 7-week progressive increasing volume exercise with high intensity. 6 hours after the last training, the rats were executed to extract the total protein of ventricular muscle for two dimensional gel electrophoresis separation. The map was analyzed with Bio-rad PDquest map analysis software. Results and Conclusion The number of protein spots on the gels of control and exercise group was 352± 17 and 338 ± 17, respectively. There were 99 protein spots expressed differentially after exercise. After exercise, 5 spots disappeared; 12 spots upgraded more than 4 folds,12 spots downgrade more than 80%; 52 spots upgraded and 42 downgraded more than 2 folds. After long-term incremental load exercise,there was significant change in ventricular muscle proteome on rat. The differential expression of protein spots were focused on 35-57 kD in molecule mass and 6.0-7.4 or 8.6-9.6 in isoelectric point. In this area, there were lots of metabolize enzyme, indicating that the change in ventricular muscle metabolism probably was the caused of the exercise induced fatigue.
