摘  要:①目的探讨c-myc在Graves disease病(GD)和乳头状甲状腺癌(PTC)患者甲状腺组织中的表达。②方法各标本均采用TRIzol法提取总RNA,RT-PCR法获取cDNA,多重PCR法同时扩增187bp的c-myc和459bp的β-actin,产物行琼脂糖凝胶电泳并照相存盘,用Gel Base/Gel Biot-Pro分析软件获取各标本c-myc和β-actin的密度值,并计算比较。③结果正常组、GD病组、PTC组的c-myc/β-actin比值(x±s)分别为:0.367±0.044、0.599±0.080和0.682±0.111。Ⅰ~Ⅱ期及Ⅲ~Ⅳ期PTC组中c-myc/β-actin的比值分别为0.596±0.077、0.740±0.092。方差分析正常组、GD病组、PTC 3组之间差异有显著性(P<0.01)。q检验结果显示:c-myc在PTC组中的表达明显高于GD病组和正常对照组,c-myc在GD病组中的表达明显高于正常对照组。t检验结果表明,在PTC标本中c-myc在Ⅲ期-Ⅳ期中的表达明显高于Ⅰ期-Ⅱ期中的表达(P<0.01)。④结论原癌基因c-myc在GD病和PTC患者甲状腺组织中的表达均增高,且随PTC分期的增加,c-myc的表达量亦随之增加。Objective To explore the expression of oncogene e - mye in thyroid tissure of patients with graves disease(GD) and papillary thyroid carcinoma(PTC). Methods Total RNA was extracted from the samples of GD,PTC tissue and thyroid normal tissue by TRIzol method, and eDNA was obtained by RT - PCR. c - myc of 187bp and β - aetin of 459bp were amplified by multiple - PCR ,the amplifieated products were separated on 1.5% agarose gel and then analyzed by GelBase/blot - Pro software of UVP system. Results The ratio of e - myc and β - actin( x^- ± s ) in normal normal control group, GD and PTC group was 0.367 ± 0.044, 0.599 ±0.080,0.682 ± 0.111 and which of stage Ⅰ - Ⅱand Ⅲ - Ⅳ in the PTC group was 0.596 ± 0. 077 and 0. 740±0. 092 respectively. There was significant difference among GD ,PTC tissue andthyroid normal control tissue( P 〈 0.01 ) by F test. The expressions of c - myc in PTC group were higher than those in GD group normal control group, and which in GD group was higher than that in normal control group by q test. The expression of c - myc in Ⅲ - Ⅳstage than that in Ⅰ - Ⅱ stage in PTC group( P 〈 0.01 ). Conclusioa The expressions of e - mye in GD and PTC were increased as the pathological stage increased.
