摘 要:采用正交试验和气质联机方法,研究了侧孢芽孢杆菌菌株BL-21降解水胺硫磷的条件和途径。结果表明:最佳碳氮源为:蔗糖1.5%,NH4HCO30.06%。最适降解温度、pH和摇床转速分别为30℃,pH 7和200 r/min。250 mL三角瓶培养基装量30 mL、接种量10%、添加10-20μL的TritonX-100有利于水胺硫磷降解。气质联机对水胺硫磷降解产物的分析表明,菌株BL-21降解水胺硫磷产生O,O’-二甲氧基硫代磷酰胺酯和水杨酸异丙酯等中间产物,O,O’-二甲氧基硫代磷酰胺酯可进一步分解成无机磷、硫化物、氨气等产物;水杨酸异丙酯降解为水杨酰胺、水杨酸及其它有机酸,进一步降解为二氧化碳和水,毒性显著降低。The degradating condition and pathway of an isocarbophos degradating bacterium, Bacillus laterosporus BL-21, were studied with orthogonal experiment and GC-MS. The results indicated that the optimum degradating ispcarbophos carbon and nitrogen sources of Bacillus laterosporus BL-21 were sucrose 1. 5% and ammonium bicarbonate 0. 06% respectively. The optimum temperature, pH and rotation speed were 30℃, pH 7 and 200 r/min respectively. While with 30 mL liquid in 250 mL flask, inoculum size 10%, and add 10%20 μL TritonX-100, strain BL-21 could degrade isoearbophos well. Using GC-MS, the intermediate products of biodegradation of isocarbophos by the strain BL-21 were detected. The results suggested that o,o’s-trimethyl phosphorothioate and isopropyl salieylate were the main intermediate products. The o,o's-trimethyl phosphorothioate was further degraded into inorganophosphate, sulfld and the ammonia. Moreover, The isopropyl salicylate was also further degraded, and created salicylamide, salicylic acid and other organic acids, at last these materials would come into being carbon dioxide and waters. Therefore, these biodegradation productions of isocarbophos by strain BL-21 were lower toxity and biological safety.
