17-β雌二醇对去卵巢大鼠小肠粘膜钙结合蛋白基因表达的影响

摘　　要：用硫氰酸胍一步法提取小肠、肾脏和骨组织总ＲＮＡ，用ＰｓｔＩ和ＳｍａＩ双酶切携带钙结合蛋白（ＣａＢＰ）９Ｋ基因的ｐＵＣ１２质粒，获取ＣａＢＰ９ＫｍＲＮＡ在各组雌性大鼠中的变化。结果：卵巢切除组，小肠粘膜ＣａＢＰ９ＫｍＲＮＡ明显低于对照组；而卵巢切除后注射１７－β雌二醇治疗组（０．５μｇ／１００ｇ体重）未出现降低。１７－β雌二醇可影响小肠粘膜ＣａＢＰ９Ｋ的基因表达。本结果提示，雌激素可能通过钙结合蛋白参与钙在大鼠体内的代谢。 Objective: To study the influence of 17βestradial on calbindin 9K gene expression in the ovariectomized rat intestines.Methods:(1)Extraction of total RNA from rat intestines kidneys and bones.(2)Expression of CaBP 9K mRNA in rat intestines by Northern blot with α 32Plabled CaBP 9K cDNA.Results: Decrease in CaBP 9K mRNA occurred in the intestines of the ovariectomized rats,but no decrease was found in those rats of the 17βestradial treatment group.Conclusion: Estrogen can influence CaBP 9K gene expression in the rat intestines.It is a possible way how estrogen regulates calcium metabolism in the rats.
