磺胺二甲嘧啶抗体的制备及酶联免疫分析方法的建立

摘  要:应用人工合成免疫原免疫动物、杂交瘤融合技术制备出抗磺胺二甲嘧啶的单抗及多抗。抗体亲和常数均大于1.0×108L/mol。利用多抗2510建立酶联免疫分析方法。该方法测量范围为0.1^30 ng/mL,灵敏度为0.08 ng/mL,批内变异〈15%,批间变异〈21%。不同样品回收率在80.5%^137.8%之间,稀释实验测定值与稀释度呈线性相关,相关系数为0.997。Polyclonal antibodies (PcAbs) and monoclonal antibodies (McAbs) against sulfamethazine (SM2) were obtained. The association constants (Ka) of the antibodies are all higher than 10^8 L/mol. The enzyme-linked immunosorbent assay (ELISA) for the measurement of SM2 was set up by using PcAb-2510. The assay range for this method is 0. 1-30 ng/mL and the sensitivity is 0.08 ng/mL. The intra-assay and inter-assay coefficients of variation (CV) vary from 12.8%-14. 1% and 14.0%-20.5%, respectively. The recovery of the analyte in different samples is from 80. 5% to 137.8% and the experimental correlation coefficient of dilution is found to be 0. 997.
