摘  要:目的探讨中药提取物神经再生素(nerve regeneration factor,NRF)对实验性急性高眼压(hyper-intraocular pressure,HIOP)兔眼视网膜神经节细胞(retinal ganglion cells,RGCs)的保护作用。方法16只健康中华大耳白兔随机等分为实验(NRF)组和空白对照(PBS)组,前房灌注法建立右眼实验性急性HIOP模型,左眼作正常对照(NC)。于灌注前、灌注后4 d、7 d,NRF组和PBS组右眼玻璃体腔内分别注射NRF4.5μg或等量(5μL)0.1 mol.L-1磷酸缓冲液。实验兔第14天安乐致死。实验兔致死前48 h以辣根过氧化物酶逆向标记双眼RGCs,视网膜铺片后以3,3’,5,5’-四甲基联苯胺法呈色,计数下半视网膜距视盘边缘为2 mm、4 mm、6 mm处的RGCs密度。结果距视盘边缘分别为2 mm、4 mm、6 mm处,NC组的RGCs密度分别为(1 621±407)mm-2、(762±235)mm-2、(366±125)mm-2;NRF组的RGCs密度分别为(1 268±378)mm-2、(699±253)mm-2、(284±104)mm-2,距视盘边缘2 mm、6 mm处,NRF组与NC组RGCs密度差异有显著统计学意义(P=0.000,0.006);PBS组的RGCs密度分别为(1 002±410)mm-2、(627±211)mm-2、(264±107)mm-2,与NC组RGCs密度差异均有统计学意义(P均<0.05);距视盘边缘2 mm处,NRF组与PBS组的RGCs密度的差异有统计学意义(P=0.033)。结论NRF可提高实验性急性HIOP兔眼RGCs的存活率,对RGCs具有保护作用。Objective To investigate the neuroprotective effect of nerve regeneration factor ( NRF), extract of Chinese herbal medicine, on retinal ganglion cells (RGCs) damaged by experimental acute hyper-intraocular pressure (HIOP). Methods Sixteen rabbits were divided randomly into experimental groups,including NRF group and blank control group (PBS). Experimental acute HIOP model was performed in the right eyes by anterior chamber perfusion of balanced salt solution,and the left eyes served as normal control group (NC). 4.5 μg of NRF or 5 μL of 0.1 mol·L^-1 PBS was injected respectively into petreous body cavity of right eyes of NRF or PBS group before perfusion and repeated on 4th, 7th day post-perfusion. All rabbits were euthanatized on 14th day. RGCs of all rabbits were antidrom-labelled with horse radish peroxidase at 48 hours before sacrifice, and colorated with 3,3', 5,5 ' -tetramethyl benzidine after retina stretched preparation. The density of RGCs was counted at inferior half retina located 2 mm,4 mm,6 mm from the optic disc. Results At the distance of 2 mm,4 mm,6 mm from optic disc,the density of RGCs of normal control group was ( 1 621 ± 407 ) mm^-2 ( 762 ± 235 ) mm^-2, ( 366 ± 125 ) mm^-2, respectively,that of NRF group was ( 1 268 ± 378) mm^-2, (699 ± 253 )mm^-2, (284 ± 104) mm^-2 ,respectively,that of PBS group was ( 1 002 ±410)mm^-2, (627 ±211 )mm^-2, (264 ± 107 )mm^- 2, respectively. At the distance of 2 mm,4 mm, 6 mm, the density of RGCs of PBS group was significantly lower than that of normal control group (P 〈 0. 05 ). At the distance of 2 mm, 6 mm, the density of RGCs of NRF group was also significantly lower than that of normal control group(P=0.000,0.006). But at the distance of 2 mm,the density of RGCs of NRF group was significantly higher than that of PBS group(P = 0.033 ). Conclusion NRF can protect RGCs from damage induced by experimental acute HIOP and enhance their survival.
