摘  要:目的观察甘草水提液对慢性肾衰大鼠肾功能的影响,探讨其抗氧化作用机制。方法Wistar大鼠30只,随机分成3组:假手术组、单纯手术组及治疗组,每组10只。检测第9周及第13周各组大鼠24 h尿蛋白、尿肌酐及丙二醛(MDA)含量;测定血中肌酐(Scr)、硒谷胱甘肽过氧化物酶(SeGSHPx)活性及晚期氧化蛋白终产物(AOPPs)的表达;计算内生肌酐清除率(Ccr);制备肾组织匀浆并检测SeGSHPx活性、MDA含量及AOPPs。结果治疗组尿蛋白排泄量、Scr均低于单纯手术组(P<0.05);治疗组Ccr及血清、肾组织匀浆中SeGSHPx活性均高于单纯手术组(P<0.05);治疗组肾组织匀浆及尿中MDA含量低于单纯手术组(P<0.05),其血清及肾组织匀浆中AOPPs也较单纯手术组低(P<0.05)。所有指标与假手术组的差异均有统计学意义(P<0.05)。结论甘草水提液对慢性肾功能不全大鼠具有保护肾功能的作用,其作用机制与抗肾脏氧化应激作用有关。Objective To investigate the effects of glycyrrhizae water-extraction on rats with chronic renal failure(CRF) and its mechanism. Methods 30 Wistar rats were randomly divided into 3 groups: the sham operation group, the simple operation group and the therapy group, 10 rats in every group. On the 9^th and 13^th weeks, the 24 h urine protein, urine creatinine and MDA levels were determined, the activities of Scr and SeGSHPx and the expression of AOPPs were determined, also the Ccr was calculated. Nephridial tissue homogenate was prepared and the activity of SeGSHPx, MDA and AOPPs were determined. Remits 24 h urine protein and Scr were obviously lower in the therapy group than those in the simple operation group( P 〈 0.05). Ccr and the activity of SeGSHPx of blood serum and nephridial tissue homogenate of the therapy group were obviously higher than those of the simple operation group ( P 〈 0.05). The MDA levels of nephridial tissue homogenate and urine in the therapy group were obviously lower than those of the simple operation group ( P 〈 0.05 ). Also, the AOPPs activities of blood serum and nephridial tissue homogenate in the therapy group were obviously lower than those of the simple operation group ( P 〈 0.05). All indexes were significantly different among the three groups. Conclusion Glycyrrhizae water-extraction provides a protective effect on renal function in rats with CRF, which is related to its antioxidation reaction.
