猪肉中克伦特罗、沙丁胺醇、土霉素、四环素残留量的HPLC同时测定方法研究

摘　　要：[目的]建立高效液相色谱法(HPLC)同时测定猪肉样品中克伦特罗、沙丁胺醇、土霉素、四环素残留量的检测方法。[方法]样品用0.01mol/LEDTA-2Na、0.3%磷酸溶液、高氯酸(1:1)混合溶液提取,上清液过C18固相萃取柱净化,采用高效液相色谱分离,二极管阵列检测器(DAD)检测,外标法定量。[结果]克伦特罗、沙丁胺醇在0.04～10μg/mL范围内线性关系良好,相关系数为0.99992～0.99993,在0.001～0.05mg/kg添加浓度范围内,平均回收率在70.3%～94.3%之间,相对标准偏差为1.52%～5.85%;土霉素、四环素在0.5～10μg/mL范围内线性关系良好,相关系数为0.99993～0.99997,在0.02～0.5mg/kg添加浓度范围内,平均回收率在64.1%～94.2%之间,相对标准偏差为1.23%～4.53%。[结论]本法简便、快速、定量准确、精密度高,适用于猪肉样品中克伦特罗、沙丁胺醇、土霉素、四环素残留量的同时检测。High performance liquid chromatographic method （HPLC） has been developed for the simultaneous determination of clenbuterol, salbutamol, oxytetracycline and tetracycline in pork. The samples were extracted with 0.01 mol/L disodium ethylenediamine tetraacetate, 0.3%phosphate, and perchloric acid（1：1）. Sample clean-up was performed on a C18 solid phase extraction column. The analytes were separated by HPLC with diode array detector and quantified using external standard calibration curves. The linear ranges of clenbuterol and salbutamol were 0.04 ~10μg/mL, and the correlation coefficients were 0.99992-0.99993. The average recoveries of clenbuterol and salbutamol in spiked pork ranged from 70.3% to 94.3%, and the relative standard deviations were between 1.52% and 5.85%. The linear ranges of oxytetracycline and tetracycline were 0.5 ~ 10μg/mL, and the correlation coefficients were 0.99993 -0.99997. The average recoveries of oxytetracycline and tetracycline in spiked pork ranged fi＇om 64.1% to 94.2%, and the relative standard deviations were between 1.23% and 4.53%. The method is simple, rapid and accurate for detection of clenbuterol, salbutamol, oxytetracycline and tetracycline in pork.
