摘 要:采用未培养技术直接从根结线虫保护地土壤样品中提取微生物总DNA,分别构建土壤细菌和放线菌的16S rRNA基因克隆文库,通过HinfⅠ和HaeⅢ限制性内切酶分别对两个基因文库中的克隆进行ARDRA分析,构建细菌和放线菌克隆文库的系统发育树,分析主要种群的组成。结果表明:根结线虫保护地土壤细菌种群主要包括α、γ、β变形细菌亚群,拟杆菌门,放线菌门等类群,其中能引起植物根癌病的根癌农杆菌所占比例较大。放线菌种群大约83.4%的克隆属于放线菌亚纲,放线菌亚目,包括微球菌亚目、小单胞菌亚目、丙酸杆菌亚目、棒状杆菌亚目和链孢囊菌亚目等,其余为未分类放线菌种类。Root-knot nematodes are obligate plant endoparasites that cause severe crop yield losses throughout the world. Community of rihizosphere microorganisms significantly influence root knot nematodes and the disease it incite. Total DNA extracted from soil infested with root-knot nematodes in protected land was used as PCR template. Community structure of bacteria and actinomyeetes were determined by anlying clone library of 16S rRNA genes amplified. The PCR products were then subeloned into pGEM-T vector. Each unique restriction fragment length polymorphism pattern, created by using two restriction endonucleases ( HinfⅠ and Hae Ⅲ ), was designated as an operational taxonomic unit (OTU). A phylogeny tree was constructed based on the ARDRA for analysing the diversity of the bacterial and actinomycetes communities. The results showed that main species of the bacterial library includes α-proteobacteria, 7-proteobaeteria, β-proteobacteria, Baeteroidetes and Aetinobacteria, et al. And Agrobacterium tumefaciens accounted for a high proportion of the bacterial library. The Actinobacteridae accounted for 83.4 % of the total Actinobacteria library. The main species belonged to the Actinobacteridae includ- ing Micrococcineae, Micromonosporineae, Propionibacterineae, Corynebacterineae, and the rests were the unclassified Actinobaeteria.
