摘 要:目的:建立以高效液相色谱法测定山楂叶中总黄酮含量的方法。方法:色谱柱为Shim-packVP-ODSC18(250mm×4.6mm,5μm),流动相为甲醇-2%醋酸溶液(30∶70),流速为1.2mL·min-1,检测波长为355nm。通过测定山楂叶主要化学成分之一金丝桃苷的含量推算出山楂叶中总黄酮的含量。结果:8份山楂叶样品中金丝桃苷与总黄酮的含量成显著正相关(r=0.98,P<0.01),且总黄酮与金丝桃苷的含量比值(换算因子)为5.955±0.300。结论:可以将金丝桃苷的含量乘以换算因子求算出不同产地、不同采收期山楂叶中总黄酮的含量。OBJECTIVE: To develop an HPLC method for determining the content of total flavonoids in leaves of Crataegus pinnatifida .METHODS:The determination was performed on Shim- pack VP -ODS C18(250 mm × 4.6 ram, 5μm) with mobile phase consisted of methanol - 2% acetic acid solution(30 : 70) at a fow rate of 1.2 mL · min ^-1.The detection wavelength was set at 355 nm.The content of total flavonoids in leaves of Crataegus pinnatifida was derived from the determination of the content of Hyperoside, one of the constituents in the leaves. RESULTS:A significant positive correlation was noted between the content of Hyperoside and the content of the total flavonoids in 8 batches of samples of leaves of Crataegus pinnatifida (the correlation coefficient was 0.98, P 〈 0.01, 2-tailed).The ratio of the total flavonoids to Hyperoside in leaves of Crataegus pinnatifida (calculation factor) was 5.955± 0.300.CONCLUSION: The total flavonoids in leaves of Crataegus pinnatifida from different production places and different harvesting time could be derived from the multiplication of the content of Hyperoside with calculation factor.
