摘 要:对纳米羟基磷灰石颗粒对成骨前体细胞MC3T3E1的增殖和分化的影响进行了研究.采用四唑盐比色法(MTT)、碱性磷酸酶检测及矿化结节染色的方法分析了羟基磷灰石纳米颗粒悬液对成骨前体细胞的增殖和分化的影响.同时用纳米羟基磷灰石悬液的浸出液及吸附条件培养液后的纳米颗粒制备悬液分别处理细胞,探讨钙离子释放和蛋白吸附在成骨前体细胞增殖和分化过程中的作用.结果表明:在0~50μg.mL-1,纳米羟基磷灰石颗粒对成骨前体细胞的增殖有一定抑制作用,且随着浓度的增加抑制作用也相应加强.10μg.mL-1的纳米羟基磷灰石悬液对成骨前体细胞的分化具有促进作用.纳米羟基磷灰石颗粒悬液对成骨前体细胞分化的促进作用不受到钙离子释放或蛋白吸附作用的影响.To study the effect of hydroxyapatite nanocrystals on proliferation and differentiation of preosteoblastic MC3T3E1 cells, MC3T3E1 cells were treated with nanocrystals in suspension culture system. MTT assay, ALP activity detection and ALP/vonkossa staining were used to analyse the effect of nanoerystals. In addition, contribution of the ion released from hydroxyapatite nanocrystals and the protein absorbed by nanocrystals for cell differentiation were tested. Hydroxyapatite nanocrystals inhibit the proliferation of MC3T3E1 cells and the inhibition increases with increasing their concentrations. The treatment of nanocrystals with a 10 μg · mL^-1 can facilitate the differentiation of MC3T3E1 cells. No positive reaction was found from the contribution of ion released from hydroxyapatite nanocrystals and protein absorbed by nanocrystals for the differentiation of MC3T3E1 cells.
