摘 要:目的:探讨中风后遗症"气虚血瘀"大鼠模型血管内皮生长因子(VEGF)的表达及补阳还五汤对其表达的影响。方法:采用机械开颅电凝法阻断大脑中动脉24h后,用随机电脉冲刺激仪每天刺激大鼠2h,连续刺激15天,复制中风后遗症"气虚血瘀"大鼠模型。治疗组在造模结束后灌服补阳还五汤水煎液,连续给药15天。通过重量法观察脑含水量的变化,并采用免疫组织化学方法和酶联免疫方法检测VEGF的表达和蛋白水平。结果:与模型组相比,补阳还五汤能降低模型大鼠脑含水量(P<0.05),VEGF阳性细胞数量明显增多(P<0.05),蛋白水平提高(P<0.05)。结论:补阳还五汤能改善中风后遗症大鼠脑水肿,并增强VEGF的表达和蛋白水平,这可能是其发挥抗脑缺血损伤作用的机制之一。Objective :To exploce the expression of vascular, endothelial growth factor (VEGF) in rats with sequelae of ischemic stroke arid effect of Buyang tiuanwu Deccoction (BYHWD) . Methods : A rat model of rats with deficiency of qi and blood stasis alter stroke was established by means of craniectomy in which the right common carotid artery was ligated with 4 - 0 silk thread followed by cautefization of the right middle cerebral, artery. Programmed electric shock was administered 24h alter the ischemic stroke for 2h daily for 15 consecutive days. The rats in sham operation group were not subjected to ligation of the fight common carotid artery or right middle cerebral artery occlusion. The rats in the treatment groups were given oral BYHWD for 15consecutive days. The expression and protein level of of VEGF in brain tissue of every groups were measured by immunohisto-ehemisty and enzyme linked immunosorbent assay. Results : The content of brain in water and the expression and protein level of VEGF were obviously different between BYHWD-treated group and model group( P 〈 0. 05). Conclusion: BYHWD can enhance the VEGF expressions and protein level in rats with deficiency of qi and blood stasis ,after stroke, which is one of possible anti-isehemie mechanism of BYHWD.
