摘  要:通过建立糖尿病小鼠的血糖波动模型,研究血糖波动对糖尿病小鼠各脏器氧化损伤以及对糖尿病并发症相关实验指标的影响.将实验小鼠分为四组,正常组小鼠、正常小鼠血糖波动组、糖尿病小鼠组、糖尿病小鼠血糖波动组.首先通过尾静脉注射四氧嘧啶5 mg/kg体重建立小鼠糖尿病模型,正常波动组和糖尿病波动组每天3次腹腔注射葡萄糖2 g/kg体重以造成小鼠的血糖波动,其余两组腹腔注射生理盐水,各组小鼠均自由进食进水.实验6周后采血,检测血清中血糖、果糖胺含量以及各脏器中丙二醛、谷胱甘肽、山梨醇等的含量.糖尿病波动组小鼠的血糖、果糖胺水平、各脏器的氧化损伤程度和山梨醇含量都显著高于糖尿病组(P<0.01).说明血糖波动能加重糖尿病小鼠的病情,并对脏器造成更为严重的损伤,这有可能是导致糖尿病并发症发生的原因之一.The effect of blcod glucose fluctuation on oxidative damage in the viscera of diabetic mice and the evaluating index of diabetes complications via the animal model-diabetes mice with blood glucose fluctuation was investigated. Mice were divided into four groups randomly: normal group, normal blood glucose fluctuation group, diabetes model group and blood glucose fluctuation with diabetes group. At first, the diabetes animal model was established by injecting alloxan through caudal vein. Then the mice in fluctuation glucose group and fluctuating glucose with diabetes group were injected with 2 g/kg body weight of glucose via abdomen so that the glucose fluctuation in blcod appears in these groups. The other groups were injected with 0.9 % of physiological salt solution via abdomen. Mice in each group were supplied enough food and water. The levels of glucose and fructosamine in serum, the contents of MDA, GSH and sorbierite in viscera were determined in all groups after six weeks. The levels of glucose and fructosamine in serum,extent of oxidative danmage and the content of sorbierite from mice in blood glucose fluctuation with diabetes group were obviously increased than that in diabetes group (P〈 0.01). The blcod glucose fluctuation aggravated the condition of diabetes in mice and damaged the vis- cera more seriously which is one of causes that induced diabetes complications.
